General description

Luminol is a very efficient and commonly used chemiluminiscent compound in condensed phase. When basic solutions of luminol containing oxygen are treated with an oxidizing agent, light is produced. Oxidising agents capable of initiating this particular reaction are Nitrogen dioxide, PAN, hydrogen peroxide, ozone and other atmospheric oxidants.

Application

  • As a chemiluminiscent substance. Used to measure opsonic and phagocytic function.
  • As a biological sensor. Used to detect the polymorphonuclear leukocytes (PMNL) response in a patient with a myeloperoxidase (MPO) deficiency.
  • Used as a forensic test for blood.
Luminol has been used in a protocol to study the immune-related responses in Arabidopsis thaliana via luminol/peroxidase-based assay.

Features and Benefits

  • Very effective chemiluminiscent compound.
  • Can detect sub-part per billion levels of metal ions with simple and inexpensive instrumentation.
Used for chemiluminescence analysis of, for example, metal cations, blood, and glucococorticoids.

When polymorphonuclear leukocytes (PMNL) and soluble or particulate matter interact, the cells produce chemiluminescence, linked to activation of the oxidative metabolism of the cells. PMNL isolated from a patient with a myeloperoxidase deficiency were found to produce almost no luminol-dependent

Luminol-dependent chemiluminescence produced by peripheral blood phagocytic cells was measured using a light detecting instrument, the Luminometer 1250 (LKB Wallac). Although less sensitive than liquid scintillation counters, the Luminometer can measure chemiluminescence effectively and detect experimentally induced and clinical variations
