General description

RFFL (Ring finger and FYVE-like domain containing E3 ubiquitin protein ligase) is a caspase-associated ring protein belonging to a family of protein ubiquitin ligases. It consists of a specialized FYVE-type zinc finger domain, a cramped phosphoinositide binding pocket and a blunted membrane insertion loop.

Application

Biochem/physiol Actions

RFFL (Ring finger and FYVE-like domain containing E3 ubiquitin protein ligase) is highly involved in the p53 signaling pathway. It directly interacts with the critical regulator of p53 turnover, MDM2 and it stabilizes MDM2 by inhibiting its ubiquitination. Its distinct FYVE domains also possess binding affinity to phosphatidylinositol 3-phosphate in lipid bilayers.
