General description

Actin, a highly conserved protein, is a major component of both the cytoskeletal and contractile structures in all cell types. It varies in amount, being related to the type of differentiation and to the func tional state of cells and tissues. Actin can be found in two different forms of aggregation, the globular or the fibrillar state, and at least six distinct isoforms occur in vertebrates. The actins exhibit over 90% sequence homology, but each isoform has a unique NH2-terminal sequence. The isoforms are comprised of three α actins (skeletal, cardiac, smooth), one β actin (β-non-muscle) and two γ actins (γ smooth muscle and γ non-muscle). Difficulties have been encountered in producing polyclonal antibodies due to the highly conserved nature of actin.
Rabbit Anti-Actin reacts with pure actin by counter immunoelectrophoresis. The product also stains the I-bands of stretched skeletal muscle.
Rabbit polyclonal anti-Actin antibody reacts with rat, chicken, human, and mouse actin. By immunoblotting, cross-reactivity with other proteins may be observed.

Application

May be used for studying the intracellular distribution of actin structures and their functions in the contractile system.
Rabbit anti-actin antibody has been used for western blot assays using mouse lung homogenates and human umbilical artery endothelial cell (HUAEC) lysates.
Rabbit anti-actin antibody was used to visualize actin in human sperm smears. Samples were incubated with the antibody (1:80) for 2 hours at 37 degrees.
Fluorescent microscopy was performed on bovine fallopian tube epithilial cells to visualize actin aggregates following inoculation with c. parvum sporozites using the rabbit anti-actin antibody from Sigma.
Rabbit polyclonal anti-Actin antibody is used to tag actin for detection and quantitation by immunocytochemical and immunohistochemical (IHC) techniques. It is used as a probe to determine the presence and roles of actin in stress fibers and I-bands of skeletal muscle cells.

Rabbit Anti-Actin is supplied as a liquid containing 0.1% sodium azide as preservative.
