General description

1-Methoxy-5-methylphenazinium methyl sulfate (MPMS) is a photochemically stable electron-transport mediator between NAD(P)H and tetrazolium dyes. The reduced form of MPMS converts tetrazolium salt to purple precipitate formazan.[1]It is a very stable and non-enzymic electron carrier that mediates electron transfer by dehydrogenases in vitro to various electron acceptors such as tetrazolium dyes or the electrode of an enzymic electric cell.

Application

1-Methoxy-5-methylphenazinium methyl sulfate has been used as a component of buffers or solutions for the assay of lactate dehydrogenase[2] and muscle succinate dehydrogenase.[3]

A synchronous enzyme-reaction system using water-soluble formazan and a non-enzymatic electron mediator was developed and applied to an enzyme immunoassay (EIA). The reaction system consists of four steps: (I) dephosphorylation of NADP(+) to produce NAD(+) by alkaline phosphatase (ALP), (II)

Valine-citrulline linkers are commonly used as enzymatically cleavable linkers for antibody-drug conjugates. While stable in human plasma, these linkers are unstable in mouse plasma due to susceptibility to an extracellular carboxylesterase. This instability often triggers premature release of drugs in
