General description

Insulin-like growth factors -1 and -2 are important regulators of signalling that mediates growth and metabolism in cells. Insulin-like growth factor -1 shares structural homology with proinsulin and is mainly produced by liver. IGF-1 acts via specific receptor, IGF-1R, a heterodimeric tyrosine kinase receptor that signals to various second messenger pathways such as MAPK, Raf/Ras and PI3K cascades. IGF 1R is fully activated by phosphorylation of tyrosine residues at Tyr1162 and Tyr1163. The effect of IGF-1 is by endocrine, paracrine and autocrine signalling. The effects of IGF-1R signalling are important at many developmental levels from cells to organ to organism, growth, differentiation, anti-apoptosis, proliferation and transformation. Anti-phospho-IR/IGF1R (pTyr1162/1163) specifically recognizes human and rat insulin receptor and insulin-like growth factor-1 receptor phosphorylated at tyrosines 1162 and 1163.

Immunogen

synthetic phosphopeptide derived from the region of IR that contains tyrosines 1162 and 1163.

Application

Anti-phospho-IR/IGF1R (pTyr1162/1163) antibody may be used for immunoblotting at a working antibody dilution of 1:1000. The antibody is suitable for immunocytochemistry.

Target description

The catalytic loops within the tyrosine kinase domains of the IR/IGF1R contain a three tyrosine motif corresponding to Tyr1158, 1162, and 1163 (for IR) and Tyr1131, 1135, and 1136 (for IGF1R). Autophosphorylation within the activation loop proceeds in a processive manner.

Physical form

Anti-phospho-IR/IGF1R (pTyr1162/1163) is supplied as a solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), pH 7.3 (+/- 0.1), 50% glycerol, with 1.0 mg/mL BSA (IgG, protease free) as a carrier and 0.05% sodium azide.
