Specificity

This specially prepared, chemically modified form of KLH is highly water-soluble and provides a large number of available carboxyl groups for conjugation to hapten. The conjugation of succinylated KLH to an amine group-containing hapten (e.g., peptide), mediated by water-soluble carbodiimide as cross-linking reagent, proceeds favorably between the newly introduced carboxyl groups of KLH and the amine groups of the hapten.

Physical form

Lyophilized with stabilizing buffer

Preparation Note

Prepared by modification of KLH with succinic anhydride.

Reconstitution

Reconstitute with 2 mL deionized water to yield an opalescent solution of KLH at 5 mg/ml in sodium phosphate buffer, pH 7.4, containing 0.32 M NaCl and 20 mM sucrose.
Reconstituted protein solution may be stored up to two months at –20°C.

In this report, we have described methods to improve the efficiency of coupling synthetic peptides to keyhole limpet hemocyanin (KLH) and for the analysis of the composition of the resulting peptide-protein conjugates. KLH was first dissolved in buffers containing 3
