Cell Line Origin

Human fibrosarcoma (retroviral packaging cells)

Cell Line Description

FLYRD18 has been developed together with FLYA13 (Sigma catalog number 95091901) for packaging of retroviruses, it is especially useful for in vivo gene delivery. Both cell lines have been derived from HT1080 (Sigma catalog number 85111505). FLYRD18 has been transfected with Moloney murine leukaemia virus cores with envelope glycoproteins of cat endogenous virus RD114. This cell line is able to produce human complement-resistant retroviral vectors at titres consistently higher than 1x10,000,000i.u./ml. Addition of 4μg/ml blasticidin S and 10μg/ml phleomycin into the media may enhance the packaging function. No replication competent virus has been detected in this cell line.

Culture Medium

DMEM + 2 mM Glutamine + 10% Fetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:20, i.e., seeding at 1x10,000 cells/cm2 using 0.05% trypsin or trypsin/EDTA; 5% CO2; 37 °C.
