Gene Information

rat ... Grm2(24415)
rhesus monkey ... Grm2(700214)

Specificity

Metabotropic glutamate receptors mGluR2 and mGluR3, which have nearly identical C-termini. There is also a very slight recognition of mGluR5. Western blot analysis of rat brain shows bands migrating at Mr = 100,000 (which contains both mGluR2 and mGluR3) and 190,000. The larger band is thought to be a dimer of the smaller band.

Immunogen

Carboxy terminus peptide of rat mGluR2 conjugated to BSA with glutaraldehyde (NGREVVDSTTSSL).

Application

Anti-Metabotropic Glutamate Receptor 2/3 Antibody detects level of Metabotropic Glutamate Receptor 2/3 & has been published & validated for use in IH(P), IH & WB.

Western blot analysis can be done at final concentrations of 0.1-0.5 μg/mL. AB1553 can be used for immunocytochemistry using paraformaldehyde or paraformaldehyde/glutaraldehyde fixed tissue with light and electron microscopy. Cryostat and vibratome sections can be used with or without Triton X-100 treatment. Final concentrations recommended are 0.1-0.5 μg/mL.

The antibody has not been tested for use in immunoprecipitation. However, antibodies to the carboxy termini of other glutamate receptors are very effective in immunoprecipitation.

Optimal working dilutions must be determined by end user.

Physical form

Affinity purified using the immunogen peptide. Lyophilized from PBS containing 1% BSA.

Storage and Stability

Maintain lyophilized material at 2-8°C or -20°C. Reconstitute to 500 μL with sterile distilled water. Store reconstituted material frozen (-20°C or -80°C) in undiluted aliquots for up to six months. Avoid repeated thawing and refreezing. Diluted antibody containing 0.1% sodium azide for Western blot analysis can be stored for several weeks and reused repeatedly.

Metabotropic glutamate receptors (mGluRs) are coupled to G protein second messenger pathways and modulate glutamate neurotransmission in the brain, where they are targeted to specific synaptic locations. As part of a strategy for defining the mechanisms for the specific targeting
Type 2 metabotropic glutamate receptor (mGluR2) fails to negatively couple to cGMP in stably transfected cells.

Metabotropic glutamate receptor 1 (mGluR1) expresses at the cell surface as disulfide-linked dimers and can be reduced to monomers with sulfhydryl reagents. To identify the dimerization domain, we transiently expressed in HEK-293 cells a truncated version of mGluR1 (RhodC-R1) devoid

Cerebellar motor coordination and cerebellar Purkinje cell synaptic function require metabotropic glutamate receptor 1 (mGluR1, Grm1). We used an unbiased proteomic approach to identify protein partners for mGluR1 in cerebellum and discovered glutamate receptor δ2 (GluRδ2, Grid2, GluΔ2) and protein
