Application

La 5-bromo-2′-desoxiuridina (5-BrdU) es un análogo de la timidina que se incorpora al ADN. La 5-BrdU se utiliza habitual y extensamente para medir la síntesis de ADN y marcar las células en división. Por consiguiente, la 5-BrdU se utiliza para estudiar la comunicación celular y otros procesos que inducen proliferación celular.
La 5-bromo-2′-desoxiuridina se ha utilizado:
  • como suplemento para estudiar el efecto de las crisis electroconvulsivas en la neurogénesis del hipocampo[1]
  • como suplemento para estudiar la pérdida de función génica en el corazón del pez cebra adulto[2]
  • en el ensayo de proliferación de linfocitos T{133]

Biochem/physiol Actions

La 5-bromo-2′-desoxiuridina (BrdU) se utiliza para analizar la proliferación celular,[3] por su fácil incorporación al ADN durante la fase S del ciclo celular. La BrdU estimula la diferenciación y la maduración celular en las líneas celulares leucémicas, mientras que inhibe la diferenciación de las células eritroleucémicas amigas.[4] En los estudios de incorporación de la BrdU, a menudo se utilizan después anticuerpos específicos de la BrdU con etiquetas fluorescentes para la detección de la BrdU incorporada, mediante métodos como la citometría de flujo y la microscopia de fluorescencia.
Análogo de timidina utilizado como mutágeno en investigación genética. Incorporada de manera selectiva en el ADN celular durante la fase S.

  1. Could specific insights into the use of this product be provided? BrdU was administered in water to mice for up to 2 weeks for an in vivo cell proliferation assay. However, upon staining with an anti-BrdU antibody, no BrdU incorporation was detected in the positive control (small intestine), despite trying various concentrations. Any information on the particular use of this product would be much appreciated

    1. Given the BrdU incorporation into DNA of cells, using the Western blot method for detection would be quite challenging. Typically, the immunohistochemistry method is employed for this purpose. When it comes to the application on small intestine samples, the following citation of B5002 via immunohistochemical detection may be useful for protocol reference:

  2. How long can BrdU be stored at -20 degrees and -4 degrees, after being diluted in sterile saline and being filtered with a 0.22 µm filter?

    1. BrdU should be stored at –20 oC and desiccated. Stock solutions can be made in DMSO to up to 50 mg/mL, as indicated in the Product information sheet. The stock can be stored for 6 months at -20°C in single use aliquots.

      No information is available on storage conditions for BrdU dissolved in saline.
