Specificity

Clone 1E10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects ZFP36L2. It targets an epitope within 17 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the internal region of human mRNA decay activator protein ZFP36L2.

Application

Quality Control Testing

Evaluated by Western Blotting in RAW264.7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ZFP36L2 in RAW264.7 cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected ZFP36L2 in Human skin tissue sections.

Affinity Binding Assay:: A representative lot of this antibody bound ZFP36L2 peptide with a KD of 9.8 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ZFP36L2 in A549 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

mRNA decay activator protein ZFP36L2 (UniProt: P47974; also known as Butyrate response factor 2, EGF-response factor 2, ERF-2, TPA-induced sequence 11d, Zinc finger protein 36, C3H1 type-like 2, ZFP36-like 2) is encoded by the ZFP36L2 (also known as ERF2, RNF162C, TIS11D) gene (Gene ID: 678) in human. ZFP36L2 is an RNA-binding protein that interacts with AU-rich elements (ARE) in the 3 untranslated region (UTR) of mRNA, which leads to mRNA degradation and translational repression. It acts as a UTR ARE mRNA-binding adapter protein to communicate signaling events to the mRNA decay machinery. It destabilizes several cytoplasmic ARE-containing mRNA transcripts by promoting their poly(A) tail removal or deadenylation, and hence provide a mechanism for attenuating protein synthesis. It is reported to shuttle between the nucleus and the cytoplasm in a XPO1/CRM1-dependent manner. Its RNA-binding domain resides in amino acids 170-211 and has two C3H1-type zinc finger domains (aa 153-181; 191-2

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
