Specificity

Clone 4G7 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Eukaryotic translation initiation factor 4 gamma 1 (eIF4G1) phosphorylated on serine 1108.

Immunogen

KLH-conjugated linear peptide corresponding to 14 amino acids surrounding phosphoserine 1108 from the internal region of human Eukaryotic translation initiation factor 4 gamma 1 (eIF4G1), isoform 3.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:100 dilution of this antibody detected phospho-eIF4G1 (Ser1108) in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:100 dilution from a representative lot detected phospho-eIF4G1 (Ser1108) in lysate from Human umbilical vein endothelial cells (HUVEC).

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected phospho-eIF4G1 (Ser1108) in PC12 cells treated with NGF.

Peptide Inhibition Assay: Target band detection in a lysate from PC12 cells treated with NGF was prevented by preblocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Eukaryotic translation initiation factor 4 gamma 1 (UniProt: Q04637-3; also known as eIF-4-gamma 1, eIF-4G 1, eIF-4G1, p220) is encoded by the EIF4G1 (also known as EIF4F, EIF4G, EIF4GI) gene (Gene ID: 1981) in human. eIF4G1 is a component of the eIF4F complex that is involved in the recognition of the mRNA cap, ATP-dependent unwinding of 5′-terminal secondary structure, and recruitment of mRNA to the ribosome. It serves as a scaffold for assembly of components of EIF4F complex that starts cap-dependent translation. This complex also plays a role in oncogenesis. Increased expression of EIF4G1 has been linked to lower median survival of patients with multiple cancers. Knockdown of eIF4G1 in prostate cancer cells is reported to reduce cyclin D1 and p-Rb protein levels and cause cell cycle delay and reduce cell viability, proliferation, and migration. eIF4G1 serves as a substrate for PKC and its phosphorylation at serine 1186 induces its binding to mitogen-activated protein kinase-interacting kinase 1 (Mnk1). Eight isoforms of eIF4G1 have been described that are produced by alternative splicing and alternative initiation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
