Specificity

Clone 1C16 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detect Rab11. It targets an epitope within 20 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal half of human Rab11.

Application

Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Rab11 in SH-SY5Y cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Rab11 in lysates from C2C12 and PC12 cells and Human brain tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Rab11 in Human stomach tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Rab11 in SH-SY5Y cells.

Affinity Binding Assay: A representative lot of this antibody bound Rab11 peptide with a KD of 1.3 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Ras-related protein Rab-11A (UniProt: P62491; also known as EC:3.6.5.2, Rab-11, YL8) is encoded by the RAB11A (also known as RAB11) gene (Gene ID: 8766) in human. The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. They cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors that are directly responsible for vesicle formation, movement, tethering and fusion. Rab11 is a lipid-anchored, evolutionarily conserved and ubiquitously expressed membrane protein that is associated with recycling endosomes and has been implicated in regulating vesicular trafficking through recycling of endosomal compartment and early endosomes to the trans- Golgi network and plasma membrane through recycling pathways. The vertebrate Rab11 consists of three members, namely Rab11a, Rab11b, and Rab25, and they have been characterized as regulators of recycling endosomes in both polarized and non-polarized cells. Rab11a acts as a major regulator of membrane delivery during cytokinesis. RAB11A is also reported to bind WD repeat domain phosphoinositide-interacting protein 2 (WIPI2) via a conserved RAB11-binding domain and regulates autophagy. Two isoforms of Rab11a have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
