Specificity

Clone 1C4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Sin3A. It targets an epitope within 23 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal region of human Sin3A.

Application

Quality Control Testing

Evaluated by Western Blotting in K562 cell nuclear extract.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected Sin3A in K562 cell nuclear extract.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Sin3A in HeLa cell nuclear Extract.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Sin3A in A431, HeLa, HepG2, and NIH 3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Sin3A in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Sin3A peptide with a KD of 2.8 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Paired amphipathic helix protein Sin3a (UniProt: Q96ST3; also known as Histone deacetylase complex subunit Sin3a, Transcriptional corepressor Sin3a) is encoded by the SIN3A gene (Gene ID: 25942) in human. Sin3a is a transcription repressor that interacts with MXI1 to repress Myc responsive genes and antagonizes Myc oncogenic activities. It also interacts with MXD1-MAX heterodimers to repress transcription by tethering SIN3A to DNA. Sin3a also serves as an important mediator of this STAT3-targeted transcriptional repression. It directly interacts with STAT3 and promotes its deacetylation. Silencing of SIN3A gene results in a prolonged nuclear retention of activated STAT3 and enhances its recruitment to the SOCS3 promoter, concomitant with histone hyperacetylation and enhanced STAT3-dependent transcription. It is also involved in the control of the circadian rhythms via transcriptional repression of circadian target genes, such as PER1. It is expressed in the developing brain, with highest levels of expression detected in the ventricular zone of various cortical regions. It is required for cortical neuron differentiation and callosal axon elongation. Mutation in SIN3A gene have been linked to Witteveen-Kolk syndrome (WITKOS), a disease characterized by global developmental delay, mild to severe intellectual disability, and facial dysmorphism. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
