Specificity

Clone 2H21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects NPM/B23 (Nucleophosmin). It targets an epitope within 20 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal region of human.

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysates.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected NPM/B23 in L6 cell lysates.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected NPM/B23 in U2OS cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected NPM/B23 in U2OS cells.

Affinity Binding Assay: A representative lot of this antibody bound NPM/B23 peptide with a KD of 5.6 x 10-7 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Various dilutions from a representative lot detected His-tagged, full-length, recombinant Human B23-NPM1 protein.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected NPM/B23 in human colon tissue sections.

Target description

Nucleophosmin (UniProt: P06748; also known as NPM, Nucleolar phosphoprotein B23, Nucleolar protein NO38, Numatrin, NPM) is encoded by the NPM1 (also known as NPM) gene (Gene ID: 4869) in human. NPM is a member a histone chaperone family that is abundant in nucleus of proliferating cells. It participates in ribosome biogenesis, mRNA processing, chromatin remodeling, embryogenesis, and regulation of tumor suppressors p53/TP53. It is generally found in the nucleolus but is translocated to the nucleoplasm following serum starvation or treatment with anticancer drugs. It is also detected in cytoplasm of subjects with primary acute myelogenous leukemia (AML), but not with secondary AML. It is reported to bind ribosome presumably to drive ribosome nuclear export. It acts as a chaperonin for the core histones H3, H2B and H4. NPM also stimulates APEX1 endonuclease activity on apurinic/apyrimidinic double-stranded DNA but inhibits APEX1 endonuclease activity on AP single-stranded RNA. It may also exert a control of APEX1 endonuclease activity within nucleoli devoted to repair AP on rDNA and the removal of oxidized rRNA molecules. It is phosphorylated on serine 4 by PLK1 and PLK2. Phosphorylation by PLK1 takes placed during mitosis and phosphorylation by PLK2 is required for centriole duplication and is sufficient to trigger centriole replication. It is also phosphorylated at serine 125 and threonine 199 by Cdk2. Phosphorylation at threonine 199 increases its affinity for ROCK2 and also triggers initiation of centrosome duplication. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.03 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
