Clone PAb416 is a ZooMAb® mouse recombinant monoclonal antibody that specifically detects Large T antigen of Simian virus 40.

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from SV40 infected COS-1 cells.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Large T antigen in lysate from SV40 infected COS-1 cells.

Tested Applications

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected SV40 Large T Antigen in COS-7 cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected SV40 Large T Antigen in one million COS-7 Cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Large T antigen (UniProt: P03070; also known as LT, LT-AG) is encoded by the SV40gp6 gene (Gene ID: 29031019) in Simian virus 40. Large T antigen is a key early protein that is reported to be essential for both driving viral replication and inducing cellular transformation. It plays a role in viral genome replication by driving entry of quiescent cells into the cell cycle and by autoregulating the synthesis of viral early mRNA. It displays highly oncogenic activities by corrupting the host cellular checkpoint mechanisms that guard cell division and the transcription, replication, and repair of DNA. It is shown to participates in the modulation of cellular gene expression preceding viral DNA replication and this step involves binding to host key cell cycle regulators retinoblastoma protein RB1/pRb and TP53. It is also known to induce the disassembly of host E2F1 transcription factors from RB1, thus promoting transcriptional activation of E2F1-regulated S-phase genes. It inhibits host TP

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
