Biochem/physiol Actions

ATP-competitive, potent and selective mTOR inhibitor.
WYE-687 is an ATP-competitive, potent and selective mTOR inhibitor (IC50 = 7 nM; [ATP] = 100 μM) with much reduced or little potency toward other kinases (PI3Kα/PI3Kγ/CK1 γ1/p38α IC50 = 0.81/3.11/17.8/28.9 μM; IC50 >50 μM toward 22 other kinases). WYE-687 selectively inhibits IGF-I-stimulated phosphorylation of S6K1 pT389 over those of AKT pS473 & pS308 in Rat1 cells (0.3 nM), as well as suppresses mTOR-dependent proliferation in cancer cultures (PC3MM2/MDA-MB-361/LNCaP/A498 IC50 = 180/200/350/620 nM).
