Application

Beas2B human lung epithelial cells were treated with TGN-020 to study the effect on cigarette smoke extracts-induced CCN1 secretion.[1]

Biochem/physiol Actions

Aquaporin 4 (AQP4) inhibitor
TGN-020 is an inhibitor of Aquaporin 4 (AQP4), the most abundant water channel in brain. Aquaporins (AQPs) are water channels required for maintaining fluid homeostasis and enabling water movement across barrier membranes, but can enhance pathological cellular volume changes and cause edema in injury states. Pretreatment with the AQP4 inhibitor TGN-020 significantly reduced the volume of brain edema associated with ischemic injury in a mouse model of focal cerebral ischemia.

Aquaporin (AQP) water channels play a significant role in mesenchymal microvascular proliferation and infiltrative growth. AQPs are highly expressed in malignant astrocytomas, and a positive correlation is observed between their expression levels and histological tumor grade. To examine the utility

The extracellular signal-regulated kinase (ERK) pathway has been reported to play a pivotal role in mediating spinal cord injury (SCI) progression. The present study aimed to investigate the effects of phosphorylated ERK1/2 (p-ERK1/2) inhibition on SCI-induced astrocyte activation and inflammation

Questions

  1. We purchased a quantity of TGN-020 earlier this year but have had exceptional difficulty finding an effective solvent for this compound (tried DCM and H2O). Does Sigma-Aldrich have data on compatible solvents? Primary lit hasn't been helpful. Thanks!

    1 answer
    1. The solubility specification for this product is >/= 8mg/mL in DMSO. The rate may be higher and is lot-specific, however, the minimum concentration in DMSO is 8 mg/mL. A brief literature search shows that HCl, Ethanol, and Dimethylformamide have also been used to dissolve this compound.
