Application

Methotrexate hydrate has been used:
  • as a dihydrofolate (DHFR) inhibitor in sphere forming assay culture[1]
  • as a component of dulbecco′ modified Eagle′s medium (DMEM) for selection of transfected cells[2][3]
  • in viability and co-culture assays [4]

Potent inhibitor of dihydrofolate reductase and agent for antitumor studies. Use to inhibit dihydrofolate reductase in DHFR-based protein expression systems. Also effective in treatment of pyrimethamine-resistant Plasmodium vivax malaria parasites.

  1. Does the powder form of this product M8407 (methotrexate hydrate) need to be stored in a desiccator?

    1. This product is the hydrate form of methotrexate. The maximum allowable water content is 12% and is reported on the lot-specific Certificate of Analysis. Dessicated storage is not necessary but is up to the end user's preference to do so. Any resulting moisture loss should be considered when preparing stock solutions. Please see the link below to review the product datasheet for A6770. This product is not tested for cell culture use, but the information contained in the document would apply to M8407.

  2. Regarding to the answer for making a solution of Methotrexate hydrate, can we use Sodium Bicarbonate to dissolve Methotrexate hydrate for cell culture?

    1. This product is not directly soluble in water and is tested for solubility in 1M Ammonium hydroxide at 50 mg/mL. This material is also soluble in 1M Sodium hydroxide. Alternatively, anhydrous DMSO may also be used to dissolve this material at 90 mg/ml. An identical Methotrexate hydrate product, A6770, has a datasheet posted to the website. This datasheet included additional information regarding general use and solution stability. Please see the link below to review this document:

    1. Methoxtrexate is soluble in 1M NaOH or NH4OH (10 mg/ml). For use in cell culture, sodium carbonate buffer (pH 9.0-9.5) can be used to dissolve methotrexate.
