General description

Mitogen-Activated Protein Kinase Kinase 4 (MEK 4 or MKK4) is a 42 kDa member of a tyrosine/threonine protein kinase family that activates the c-Jun NH2-terminal kinases (JNK), which is part of the inflammation/stress signaling pathway. The mutation of the MEK 4 gene in some carcinomas indicates that it may also have a role as a tumor suppressor. Phosphorylation of MEK4 by MEKK1 on serine 257 and threonine 261 in the catalytic domain activates the protein and enables MEK 4 to phosphorylate JNK
This antibody specifically recognizes human MEK 4 double phosphorylated at serine 257 and threonine 261. Mouse and frog MEK 4 (100% homology) may cross react.

Immunogen

synthetic MEK4-derived phosphopeptide that is phosphorylated at serine 257 and threonine 261.

Application

Anti-phospho-MEK4 (Tyr397) antibody may be used for detection by immunoblotting at a working concentration of 0.1-1.0 μg/mL.

Physical form

Solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), pH 7.3, with 1.0 mg/mL BSA (IgG and protease free) and 0.05% sodium azide.

The Mitogen-Activated Protein Kinase Kinase 4 (MKK4), a member of the MAP kinase kinase family, directly phosphorylates and activates the c-Jun NH2-terminal kinases (JNK), in response to cellular stresses and proinflammatory cytokines. JNK is a member of the MAP kinase

MEK kinases (MEKKs) comprise a family of related serine-threonine protein kinases that regulate mitogen-activated protein kinase (MAPK) signalling pathways leading to c-Jun NH2-terminal kinase (JNK) and p38 activation, induced by cellular stress (e.g., UV and gamma irradiation, osmotic stress, heat

The c-Jun N-terminal kinase (JNK) group of mitogen-activated protein kinases (MAP kinases) is activated by exposure of cells to environmental stress and by the treatment of cells with cytokines. The mechanism of activation of JNK is mediated by dual phosphorylation
