General description

Interleukin-10 (IL-10) is a pleotropic cytokine with anti-inflammatory and immunosuppressive functions. It is the key cytokine that regulates the host inflammatory responses to bacterial, fungal and viral pathogens. The production of IL-10 is mediated by signaling of IL-6, IL-12 and TGF-β. IL-10 is secreted by a variety of cells including T cells, B cells, dendritic cells, eosinophils, natural killer cells and mast cells. The effects of IL-10 are mediated by binding to the IL-10R, consisting of α and β subunits. The signaling through IL-10R activates Jak1, Tyk2 protein kinases and JAK/STAT3 pathways. IL-10 is involved in suppression of innate immunity to kill pathogens and inhibits the release of reactive oxygen and nitrogen species from activated macrophages. Regulation of IL-10 production is important in infections, inflammatory responses in autoimmune encephalomyelitis, endotoxic shock and chronic enterocolitis. Anti-interleukin-10 antibody is specific for human IL-10.

Specificity

This antibody will neutralize the biological activity of recombinant human IL-10.

Immunogen

recombinant human IL-10.

Application

Anti-interleukin-10 antibody may be used for immunoblotting at a working concentration of 1-2 μg/ml. The antibody is suitable for neutralization assays at a minimum antibody concentration of 2 μg/ml.

Physical form

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline, pH 7.4, containing 5% trehalose.

Preparation Note

purified using Protein A or G

IL-10 plays an essential part in controlling inflammation and instructing adaptive immune responses. Consequently, dysregulation of IL-10 is linked with susceptibility to numerous infectious and autoimmune diseases in mouse models and in humans. It has become increasingly clear that appropriate

The generation of an effective immune response against an infection while also limiting tissue damage requires a delicate balance between pro- and anti-inflammatory responses. Interleukin-10 (IL-10) has potent immunosuppressive effects and is essential for regulation of immune responses. However, the

Interleukin-10 (IL-10), first recognized for its ability to inhibit activation and effector function of T cells, monocytes, and macrophages, is a multifunctional cytokine with diverse effects on most hemopoietic cell types. The principal routine function of IL-10 appears to be
