Porcine thyroid epithelial

Cell Line Description

The cell line was spontaneously derived from the porcine thyroid cell line PORTHOS (Sigma Catalogue number. 93122323) after withdrawal of thyroid stimulating hormone (TSH). The cells form follicle-like structures, have a high growth rate, a high glucose consumption and are TSH independent for growth. The expression of plasminogen activators has been reported and found to be higher in ATHOS cells compared to PORTHOS cells.

Culture Medium

Click:RPMI (1:1) + 10μg/ml Insulin + 10nM Hydrocortisone + 5μg/ml Transferrin + 10ng/ml glycyl-L-histydyl-L-lysine acetate + 10ng/ml somatostatin + 2mM Glutamine + 0.1% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 i.e. seeding at 5x100,000 cells/cm2 using 0.05% trypsin/0.025% EDTA in Puck F saline; 2.5% CO2; 37°C.
