The cell line CL21 has been established by hybridisation of C57 BL mouse peritoneal macrophages with SV40 transformed human skin fibroblasts. The human cells were derived from a patient with Lesch-Nyhan syndrome and are deficient in hypoxanthine guanine phosphoribosyltransferase (HGPRT). They have been reported to contain the human chromosome 7 and could also contain subchromosomal fragments of chromosome 17.
Split sub-confluent cultures (70-80%) 1:3 to 1:6 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
