General description
Llama, camels, and alpacas are camelids that produce normal IgG1 like antibodies and also functional IgG isotypes that do not incorporate light chains and possess only heavy chains. These heavy-chain antibodies may constitute about 50% of the IgG in llama serum and as much as 75% of the IgG in camel serum. By virtue of their smaller size they exhibit better bio-distribution, tissue penetration, and monomeric behavior. IgG3 consists of dimers of short heavy chains, which are characterized by a normal Fc region without a CH1 domain. Although it lacks the CH1 region, it still retains an antigen binding domain called the VHH region. Clone 8E1 is shown to specifically bind to IgG3 and not to any of the IgG2 heavy chains. (Ref.: Daley, LP., et al. (2005). Clin. Diag. Lab. Immunol. 12(4); 380-386).
Specificity
Clone 8E1 is a mouse monoclonal antibody that detects Llama IgG3.
Immunogen
Purified Llama serum IgG3.
Application
Quality Control Testing
Evaluated by Western Blotting with IgG3.
Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Llama IgG3, but not IgG1 and IgG2.
Tested Applications
Immunoaffinity Purification: A representative lot was used in Immunoaffinity Purification applications (Daley, L.P., et. al. (2005). Clin Diagn Lab Immunol. 12(3):380-6; Daley-Bauer, L.P., et. al. (2010). Clin Vaccine Immunol. 17(12):2007-15).
ELISA Analysis: A representative lot detected Mouse Anti-Llama IgG3 in ELISA applications (Daley, L.P., et. al. (2005). Clin Diagn Lab Immunol. 12(3):380-6; Daley-Bauer, L.P., et. al. (2010). Clin Vaccine Immunol. 17(12):2007-15).
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
