General description

STAM-binding protein (STAMBP) is a critical metalloprotease in ubiquitin-mediated signaling events that specifically cleaves ′Lys-63′-linked polyubiquitin chains but does not cleave ′Lys-48′-linked polyubiquitin chains. STAM-binding protein is a deubiquitinating (DUB) isopeptidase also known as AMSH, for associated molecule with the SH3 domain of STAM. STAM-binding protein plays a role in cell growth and MYC induction, potentiates BMP signaling and is key in regulating the cell surface receptor-mediated endocytosis and ubiquitin-dependent sorting of receptors to lysosomes. STAM-binding protein interacts with other key proteins including STAM1, SMAD6/7, SMURF2, and CHMP3. STAM-binding protein is widely expressed and can be found localized to the nucleus, cytoplasm and endosome. Defects in STAM-binding protein result in a congenital syndrome named Microcephaly-capillary malformation syndrome.
