General description

Serine/threonine-protein phosphatase PP1-alpha catalytic subunit (PP-1A) encoded by the gene named PPP1CA or PPP1A is one of the three catalytic subunits of protein phosphatase 1 (PP1), one of the major protein phosphatases in the cell. PPP1CA interacts with over 200 regulatory proteins to form highly specific holoenzymes which dephosphorylate a huge variety of target proteins. PPP1CA is critical for cell division, chromatin structure, protein synthesis, glycogen metabolism, muscle and cardiac function as well as the regulation of long-term ionic conductances and even synaptic plasticity. Developmentally it has nearly as many roles with principal ones being as a regulator of neural tube and optic fissure closure as well as enteric neural crest cell (ENCCs) migration during development. PPP1CA can be found both cytoplasmically and nuclear localized depending upon its regulatory partner but PPP1CA is mostly nuclear/nucleoplasm localized. Interestingly in the brain, PPP1CA also functions as a suppressor of learning and memory and as a potential mediator of cognitive decline during aging. EMD-Millipore’s Anti-PPP1CA mouse monoclonal antibody has been tested in western blot on recombinant protein as well as cell lysates from HeLa, HepG2, MCF-7, Jurkat and A549 cells and by direct ELISA against varying concentrations of antigen.

Evaluated by Western Blotting in HeLa, HepG2, MCF-7, Jurkat, and A549 cell lysates.

Western Blotting Analysis: A 1:500-2,000 dilution of this antibody detected PPP1CA in HeLa, HepG2, MCF-7, Jurkat, and A549 cell lysates.

Optimal working dilutions must be determined by end user.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
