Gene Information

General description

Killer cell lectin-like receptor subfamily B member 1B allele B (UniProt: Q5NKN4; also known as CD161 antigen-like family member B, Immunoreceptor NKR-P1C, Natural killer cell surface protein NKR-P1B allele WAG/PVG/BS, Natural killer lymphocyte receptor P1B, CD161b) is encoded by the Klrb1b (also known as Nkrp1b, Nkrp1c) gene (Gene ID: 678513) in rat. NKR-P1B is a single-pass type II membrane protein that is expressed in a subset of natural killer cells. It consists of a cytoplasmic domain (aa 1-45), a helical domain (aa 46-66), and an extracellular domain (aa 67-223). In contrast to NKR-P1C, which functions as an activating receptor, NKR-P1B inhibits NK cell activation. NKR-P1b serves as a receptor for CLEC2D/OCIL and ligand binding contributes to inhibition of cytotoxic natural killer cells. It may also mediate MHC class I-independent ′missing-self′ recognition of allografts, tumor cells, and virus-infected cells. The inhibitory NKR-P1B receptor identifies rat splenic NK cells that are low in Ly49 receptors, but are enriched in CD94/NKG2 receptors. (Ref.: Inngjerdingen, M et al. (2012). J. Immunol. 188(6); 2499-2508; Carlyle, JR et al. (1999). J. Immunol. 162(10); 5917-5923).

Specificity

Clone Stok27 is a rat monoclonal antibody that detects Killer cell lectin-like receptor subfamily B member 1B allele B (NKR-P1B) in rat. It targets an epitope within the extracellular domain.

Immunogen

IL-2-activated NK cells from PVG.1N (RT1n) rats.

Application

Anti-NKR-P1B (CD161b), clone Stok27, Cat. No. MABF1971, is a rat monoclonal antibody that detects NKR-P1B and has been tested for use in Flow Cytometry and Immunoprecipitation.
Immunoprecipitation Analysis: A representative lot detected NKR-P1B (CD161b) in Immunoprecipitation applications (Kveberg, L., et. al. (2006). J Immunol. 176(7):4133-40).

Flow Cytometry Analysis: A representative lot detected NKR-P1B (CD161b) in Flow Cytometry applications (Kveberg, L., et. al. (2006). J Immunol. 176(7):4133-40; Smelt, M.J., et. al. (2014). Cell Transplant. 23(11):1381-94).

Quality

Evaluated by Flow Cytometry in Rnk16 (Rat NK) cells stably transfected with Stok27 (NKR-1B) antigen.
