Application

Quality Control Testing

Evaluated by Flow Cytometry in C57BL/6 Mouse splenocytes.

Flow Cytometry Analysis: 1.0 µg of this antibody detected IFNGR1/CD119 in one million C57BL/6 Mouse splenocytes.

Tested Applications

Flow Cytometry Analysis: A representative lot detected IFNGR1/CD119 in Flow Cytometry applications (Haring, J.S., et al. (2005). J Immunol. 174(11):6791-802).

Target description

Interferon gamma receptor 1 (UniProt: P15261; also known as IFN-gamma receptor 1, IFN-gamma-R1, Interferon gamma receptor alpha-chain, IFN-gamma-R-alpha, CD119) is encoded by the Ifngr1 (also known as Ifngr) gene (Gene ID: 15979) in murine species. IFNGR1/CD119 is single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 26-254), a transmembrane domain (aa 255-275), and a cytoplasmic domain (aa 276-477). It serves as a receptor subunit for interferon- that plays critical roles in antibacterial, antiviral, and antitumor responses by activating effector immune cells and enhancing antigen presentation. It associates with IFNGR2 to form a functional receptor. Upon ligand binding, the intracellular domain of IFNGR1/CD119 opens out to allow association of downstream signaling components JAK1 and JAK2. In turn, the activated JAK1 phosphorylates IFNGR1/CD119 to form a docking site for STAT1. Subsequent phosphorylation of STAT1 leads to its dimerization, translocation to the nucleus, and stimulation of target gene transcription. Phosphorylation of the intracellular domain of IFNGR1/CD119 also provides a docking site for SOCS1 that regulates the JAK-STAT pathway by competing with STAT1 binding to IFNGR1.
