General description

Chloride intracellular channel protein 4 (UniProt: Q9XSA7; also known as Intracellular chloride ion channel protein p64H1, CLIC4) is encoded by the CLIC4 gene (Gene ID: 286823) in bovine. CLIC4, a member of the CLIC family of proteins. CLIC family members can change from a globular, soluble state to a state where the N-terminal domain is inserted into the membrane and functions as chloride channel. A conformation change of the N-terminal domain is thought to expose hydrophobic surfaces that trigger membrane insertion. CLIC4 is ubiquitously expressed and is present in both soluble and membrane-associated forms. It is present in the cytosol as a diffusible protein, but is reported in endoplasmic reticulum, trans-Golgi network, mitochondria, caveolae, dense-core secretory vesicles in the central nervous system, and in nuclei of differentiating keratinocytes. CLIC4 is rapidly recruited to the plasma membrane by lysophosphatidic acid (LPA) and serum, indicating its possible role in exocytic endocytic trafficking. CLIC4 also participates in such diverse processes as angiogenesis, differentiation, migration and wound healing. It is reported to stimulate both the internalization and serum/ LPA-induced recycling of beta1 integrin. In developing and developed kidneys, CLIC4 is enriched in the proximal tubule epithelial cells where it is considered to be essential for luminal delivery, microvillus morphogenesis, and endo-lysosomal biogenesis. CLIC4-null mice embryos exhibit impaired renal tubulogenesis and have smaller kidneys with fewer glomeruli and less dense peritubular capillary network. (Ref.: Edwards, JC et al. (2014).
