General description

Cubilin (UniProt O60494; also known as 460 kDa receptor, Intestinal intrinsic factor receptor, Intrinsic factor-cobalamin receptor, Intrinsic factor-vitamin B12 receptor) is encoded by the CUBN (also known as IFCR, MGA1) gene (Gene ID 8029) in human. Cubilin is a large 460 kDa glycosylated extracellular protein composed of seven EGF repeats (a.a. 132-468) and twenty seven CUB (complement C1r/C1s, Uegf [epidermal growth factor related sea urchin protein], and bone morphogenic protein 1) domains (a.a. 474-3623). Cubulin binds ligands via its CUB domains and mediates ligands endocytosis via interaction with other membrane proteins. Cubulin operates in concert with megalin n the proximal tubule and mediates the reabsorption of proteins filtered in the glomeruli. Cubulin also interacts, via its EGF domains, with the transmembrane protein amnionless (AMN), neither protein can be localized at the membrane without the presence of the other. Mutations in either the CUBN or the AMN gene are linked to the rare autosomal recessive disorder known as Imerslund-Gräsbeck syndrome (IGS) or megaloblastic anemia 1 (MGA1), characterized by B12 deficiency due to impaired intestinal B12 absorption. Cubilin is synthesized as a 3623-a.a. precursor wtih a signal peptide (human a.a. 1-23, rat a.a. 1-20) and a propeptide (human a.a. 24-35, rat a.a. 21-32) sequence, which are removed posttranslationally to yield the mature protein.

Specificity

This polyclonal antibody targets the mature cubilin N-terminal end region prior to the first EGF domain.

GST-tagged recombinant human mature cubilin N-terminal end fragment.

Application

Detect Cubilin using this rabbit polyclonal Anti-Cubilin Antibody, Cat. No. ABS1070, validated for use in Immunohistochemistry (Paraffin) and Western Blotting.
Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected cubilin in rat testis, human kidney and small intestine tissue sections.

Target description

Greater than 420 kDa observed. Target band size appears larger than the calculated molecular weights of 398.7/396.0/394.7 kDa (human prepro-/pro-/mature form) and 399.0/396.7/395.4 kDa (rat prepro-/pro-/mature form) due to glycosylation. Uncharacterized bands may be observed in some lysate(s).
