General description

CARM1, also known as Histone-arginine methyltransferase CARM1, Coactivator-associated arginine methyltransferase 1, Protein arginine N-methyltransferase 4, and encoded by the gene name CARM1 and PRMT4, catalyzes the transfer of a methyl group from S-adenosyl-L-methionine to arginine residues. CARM1 methylates (mono- and asymmetric dimethylation) the the guanidino nitrogens of arginyl residues in several proteins involved in DNA packaging, transcription regulation, pre-mRNA splicing, and mRNA stability. Upon gene activation CARM1 together with histone acetyltransferases from EP300/P300 and p160 families, methylates histone H3 at ′Arg-17′ (H3R17me), forming mainly asymmetric dimethylarginine (H3R17me2a), leading to activate transcription via chromatin remodeling. During myogenic transcriptional activation, acts together with NCOA3/ACTR as a coactivator for MEF2C. During monocyte inflammatory stimulation, acts together with EP300/P300 as a coactivator for NF-kappa-B. Acts as coactivator for PPARG, promotes adipocyte differentiation and the accumulation of brown fat tissue. Plays a role in the regulation of pre-mRNA alternative splicing by methylation of splicing factors. Also methylates arginine residues in RNA-binding proteins PABPC1, ELAVL1 and ELAV4, which may affect their mRNA-stabilizing properties and the half-life of their target mRNAs. CARM1 is overexpressed in prostate adenocarcinomas and high-grade prostatic intraepithelial neoplasia.

Application

Immunoprecipitation Analysis: A representative lot immunoprecipitated methyl-CARM1 (Arg551) in vitro methylation of a control buffer, 15 mg nuclear lysates, and 15 mg cytoplasmic lysates from CARM1/MEF cells by 1 mg CARM1 WT and CARM1 R551K (Kuhn, P., et al. (2011). Nucleic Acids Research. 39(7):2717-2726).

Quality

Evaluated by Western Blotting in HEK293T CARM1 KO cells restored with WT CARM1, but not in HEK293T CARM1 KO cells restored with R551K mutant CARM1 cell lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected methyl-CARM1 (Arg551) in 10 µg of HEK293T CARM1 KO cells restored with WT CARM1, but not in HEK293T CARM1 KO cells restored with R551K mutant CARM1 cell lysate.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
