Specificity

Specific for Adenosine A3 receptor. Based on mRNA studies, the A3R has been found in rat testis and lung, kidneys, heart and brain. Other studies have found the mRNA to be highest in lung, spleen and pineal glands and moderate levels in brain kidney and testis.

Immunogen

Amino acid sequence corresponding to the 3rd extracellular domain (15 amino acids) of the rat A3R gene.

Western blot: 1-10 micrograms per mL: A3 receptors are different sizes in different species & tissues; A3R from rat brain membranes runs as a 40kDa band, 52kDa in rabbit uroepithelium membranes (Yu, W et al. Am J. Physiol Cell Physiol 2006 in press); use proteinase inhibitors and membrane preparations for best signals.

ELISA: 1:10,000-1:50,000 (50-100 ng A3R peptide (Catalog Number AG294)/well)

Immunocytochemistry: Caufield, 2003. Light 2% PFA fixation 5-10 minutes --long fixations lower staining intensity.

Optimal working dilutions must be determined by end user.

Physical form

Affinity purified immunoglobulin. Liquid in PBS with 0.1% BSA

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 12 months. Avoid repeated freeze/thaw cycles.
