General description

A concentrate of dithiothreitol (DTT) in phosphate buffer. Useful for the isolation of pathogenic or saprophytic bacteria, fungi, yeast, and epithelial cells from sputum and other mucoid fluids.
Concentrate of Dithiothreitol (DTT; Cat. No. 233155) in phosphate buffer, pH 6.5-7.5. Intended for the isolation of epithelial cells, pathogenic or saprophytic bacteria, fungi, and yeasts from sputum. Each vial is sufficient to make 100 ml of working solution.
Note: 1 set = 20 x 10 ml.

Physical form

20 vials each containing 10 ml of concentrated DTT OR 1 vial containing 10 ml of concentrated DTT.

Preparation Note

Dilute the contents of one vial to 100 ml with sterile distilled water. Swirl gently to dissolve any crystals. If the diluted material appears cloudy, the solution may be filtered to remove any insoluble particles. The diluted solution is ~6.5 mM dithiothreitol in 100 mM phosphate buffer, pH 7.0.

Reconstitution

Once a vial is opened the unused concentrate should be purged with an inert gas (e.g. N₂) and refrigerated (4°C). Diluted solutions, stored aseptically under an inert gas, are stable for up to 48 h at 4°C.

    1. The presence of DTT in 560000 may be inhibiting signal detection by quenching the fluorescence reading from PCR. It is advisable to remove DTT before conducting PCR analysis.
