General description
Anti-HNE-Michael Adducts, Reduced, rabbit polyclonal, specifically recognizes chemically reduced amino acid-(4)-HNE adducts. It is validated for use in ELISA, WB & IHC (frozen & paraffin sections).
Rabbit polyclonal antibody supplied as diluted serum. Recognizes, specifically, chemically reduced amino acid-(4)-HNE adducts.
Recognizes chemically reduced amino acid-(4)-HNE adducts. Does not recognize non-reduced adducts or reduced modifications arising from malondialdehyde (MDA) and structurally-related alkenes such as trans-2-noneal.
The CNS is enriched in polyunsaturated fatty acids and is therefore particularly vulnerable to lipid peroxidation. 4-Hydroxy-2-noneal (HNE) is one of the major lipid peroxidation products with cytotoxic and mutagenic activity. It reacts with protein residues such as histidine to generate stable Michael adducts. HNE has been shown to inhibit neurite outgrowth and disrupt microtubules and tubulin modification, suggesting it may play a role in Alzheimer′s disease.

Immunoblotting (1:1000-1:4000)
Frozen Sections (1:50-1:500)
Paraffin Sections (1:50-1:500)
Warning
Toxicity: Standard Handling (A)
Physical form
Serum diluted in 50% glycerol, pH 7.5.
Reconstitution
Following initial thaw, aliquot and freeze (-70°C).
Other Notes
Does not bind non-reduced adducts and reduced modifications arising from malondialdehyde (MDA) or structurally-related alkenals such as trans-2-noneal. Does not cross-react with non-reduced amino acid-HNE Michael adducts. Antibody binding requires the presence of the 4-hydroxyl group. Variables associated with assay conditions will dictate the optimal working dilution.
