General description

Dual specificity mitogen-activated protein kinase kinase 7 (UniProt: O14733; also known as EC:2.7.12.2, MAP kinase kinase 7, MAPKK 7, JNK-activating kinase 2, MAPK/ERK kinase 7, MEK 7;Stress-activated protein kinase kinase 4, SKK4, SAPK kinase 4, SAPKK-4, SAPKK4, c-Jun N-terminal kinase kinase 2,JNK kinase 2, JNKK 2) is encoded by the MAP2K7 (also known as JNKK2, MEK7, MKK7, RKMK7, SKK4) gene (Gene ID: 5609) in human. MKK4/SKK4 belongs to the STE Ser/Thr protein kinase family and is an essential component of the MAP kinase signal transduction pathway. MKK4/SKK4 is ubiquitously expressed with highest level of expression reported in skeletal muscle. It contains a single protein kinase domain and has a DVD domain (aa 377-400) that contains a conserved docking site and bind to their specific upstream MAP kinase kinase kinases (MAPK3Ks). MKK4/SKK4 also has a D domain in the N-terminal region (aa 37-57), which contains a conserved docking site that is required for the binding of MAPK substrates. It is activated by phosphorylation on Ser271 and Thr275 by MAP kinase kinase kinases (MAP3Ks) and it directly phosphorylates and activates the c-Jun NH2-terminal kinases (JNK), in response to cellular stresses and pro-inflammatory cytokines. Mice lacking the MKK4 gene are shown to have abnormal hepatogenesis and die before embryonic day 14.

Specificity

This rabbit polyclonal antibody detects MKK7/SKK4 phosphorylated on Thr275/Ser277.

Immunogen

peptide corresponding to amino acids surrounding Thr275/Ser277 of human MKK7/SKK4.

Anti-Phospho-MKK7/SKK4 (Thr275/Ser277), Cat. No. 36-010, is a highly specific rabbit polyclonal antibody that detect dual specificity mitogen-activated protein kinase kinase 7 (MKK7) and has been tested for use in Western Blotting.

Quality

Evaluated by Western Blotting in lysate of A431 cells treates with Calyculin A and okadaic acid.

Western Blotting Analysis: A 1:500 dilution of this antibody detected Phospho-MKK7/SKK4 (Thr275/Ser277) in lysate from 10 µg of A431 cells treated with Calyculin A and okadaic acid.
