General description

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci.

Specificity

Broad species cross-reactivity is expected.
Specificity of this polyclonal antibody toward histone H3 Lys4 dimethylation was demonstrated by dot blot analysis using the unconjugated antiserum (Cat. No. 07-030).

Epitope: H3K4me2
KLH-conjugated synthetic peptide (ART[Kme2]QTAR-GC) corresponding to amino acids 1-8 of human histone H3.

The unconjugated antiserum (Cat. No. 07-030) is shown to be suitable also for dot blot, chromatin immunoprecipitation (ChIP), ChIP-seq, immunocytochemistry, immunofluorescence, and Western blotting applications.
This Anti-dimethyl Histone H3 Antibody (Lys4), Alexa Fluor 647 Conjugate is validated for use in Immunocytochemistry for the detection of dimethyl Histone H3.

Quality

Evaluated by Immunocytochemistry in A431 cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected nuclear histone H3 Lys4 dimethylation in A431 cells.
