General description

(2-Hydroxypropyl)-β-cyclodextrin, a hydroxyalkyl derivative, finds application as a formulation vehicle that is used to enhance the solubility of the drugs in aqueous solutions for the successful delivery of medical agents to biological systems. It is generally employed as a substitute for α-, β- and γ-cyclodextrins.[1]

Application

(2-Hydroxypropyl)-β-cyclodextrin may be used:
  • As an analytical standard for the determination of the analyte in pharmaceutical formulations by thin layer chromatography (TLC) method.[2]
  • To study the host-guest interaction with organic low molecular mass compounds prior to their quantification using reversed phase-high performance liquid chromatography (RP-HPLC) technique.[3]
  • As a mobile phase additive for the effective separation of β-carbolines by HPLC.[4]
  • In cyclodextrin-modified micellar electrokinetic chromatography (CD-modified MEKC) for the determination of sertraline hydrochloride and the synthesis-related products.[5]

The solubility of lipophilic drugs increases linearly with the concentration of hydroxypropyl-β-cyclodextrin (HP-β-CD) in aqueous solution because of the complex between HP-β-CD and the drug. This guest-host type complex is formed between the drug and the non-polar cavity in the HP-β-CD that results in enhanced solubility.[1][6] HP-β-CD can be used to prepare an inclusion complex with ropivacaine (RVC) to increase the solubility of RVC for efficient drug delivery.[7]
The solubility of lipophilic drugs increases linearly with the concentration of hydroxypropyl-β-cyclodextrin (HBC) in aqueous solution because of the complex between HBC and the drug. This guest-host type complex is formed between the drug and the non-polar cavity in the HBC that results in enhanced solubility. Solutions may be lyophilized to produce freely soluble powders. Non-toxic in rabbits and mice.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. I purchased this material which lists the "molar substitution" as 0.8-1.0 with a MW of ~1460. The C of A specifies the molar substitution as 0.9, does this correspond to MW of 1500? And what is the distribution of MW?

    1. Yes, the molar substitution is used to determine the 1500 MW. The following example calculations may be helpful:
      Molecular Substitution = (1460 -1135.12) / 7 / 58.09 09 = 0.80 (# of substituted units per ring)
      Where 1460 is the average molecular weight
      Where 1135.12 is the molecular weight of the material if it had no substitution (if all R were H and not C3OH7)
      Where 7 is the number of repeating glucopyranose units for this compound
      Where 58.09 is the molecular weight of one substitution - C3OH7
