一般說明

促肾上腺皮质激素释放因子(CRF),一种41个氨基酸的神经肽,属于尿皮质素(urocortin)、蛙皮降压肽(sauvagine)和尾加压素I等结构相关肽组成的家族。CRF由196个氨基酸的较大前体蛋白(preproCRF)加工而来。CRF广泛分布于大脑和周围神经系统,在下丘脑室旁核(PVN)中浓度最高。

特異性

抗体不与人或大鼠CRF(6-33)、羊CRF、大鼠尿皮质素、PACAP38、人ACTH、人ACTH(18-39)和蛙皮降压肽交叉反应。

免疫原

KLH标记的合成肽,对应人CRF蛋白的C端区域(氨基酸25-41,N-末端加入赖氨酸)。

應用

兔抗促肾上腺皮质激素释放因子抗体可用于免疫组织化学(1:2000)和放射免疫分析。

生化/生理作用

促肾上腺皮质激素释放因子(CRF)是下丘脑-垂体-肾上腺轴及自主和行为应激反应的重要调节因子。CRF效应包括改变运动和认知行为、控制食欲和调节免疫系统。CRF是体外和体内垂体前叶促肾上腺皮质激素细胞分泌的促肾上腺皮质激素(ACTH)的强效刺激因子。CRF效应通过CRF受体亚型CRFR1和CRFR2(剪接变体)介导。CRF高亲和性结合CRF结合蛋白(CRF-BP),导致CRF失活,可将CRF从CRF-BP解离出来的配体具有认知增强作用。CRF过量与厌食和抑郁有关,可能会导致库欣综合征。CRF水平与阿尔茨海默症(AD)的致病过程相关。

外觀

溶于0.01 M磷酸盐缓冲液(pH 7.4)中,含有15 mM叠氮化钠。

儲存和穩定性

如需连续使用,可在2-8℃保存至多一个月。 若需延长储存时间,可将溶液分装并冷冻。 不建议反复冻融。 不建议在无霜冰箱中储存。 如果长期储存后发生轻微浑浊,请在使用前通过离心澄清溶液。
