一般說明

MMPs(基质金属蛋白酶)是负责细胞外基质(ECM)分解的锌蛋白酶家族,在正常的生理过程中起关键作用,例如胚胎发育和组织形态发生、组织和骨骼重塑、伤口愈合和血管生成。这些过程依赖于MMPs在细胞表面受体的裂解、凋亡配体的释放、细胞增殖和分化以及趋化因子活性调节中的作用。在结构上相似,MMP被合成并分泌为需要蛋白水解切割才能激活的无活性原酶。该过程可以由丝氨酸蛋白酶或其他MMP介导。MMP表达的增加是对多种刺激(包括粘附分子、生长因子、细胞因子和激素)的响应而发生的。MMP活性的调节主要由TIMP(金属蛋白酶的组织抑制剂)控制。因此,MMP/TIMP平衡的破坏可导致自身免疫性疾病、关节炎、心血管疾病以及肿瘤的生长和转移。
