General description

琼脂糖溶液为即用型,可用于免疫沉淀。使用15µ l混悬剂/µg一抗。推荐将蛋白A-琼脂糖用于使用兔多克隆抗体或未修饰的小鼠IgG2a或IgG2b单克隆抗体的免疫沉淀测定。小鼠IgG1或IgG3和全部大鼠抗体与蛋白A的结合较弱。作为替代方案,使用蛋白G PLUS,蛋白G PLUS/蛋白A或兔第二步抗体将这些抗体与蛋白A桥接。预清除将使由非特异性沉淀产生额外的条带最小化。要进行预清除,将与免疫沉淀抗体相同物种的 20 µl 琼脂糖结合物和 1 µg 正常 IgG 加入样品中。当使用免疫印迹进行检测时,某些二抗可以与BSA或样品中存在的其他高浓度蛋白质发生非特异性反应。这可以通过降低二抗的浓度来消除。
