General description

ZAP-70 is a protein tyrosine kinase that is expressed in natural killer cells and T-lymphocytes. ZAP-70 interacts with the ζ chain of the T cell antigen receptor (TCR) and subsequently modulates cell signaling. Phosphorylation of Tyr315 and Tyr319 residues regulates TCR-mediated signal transduction . Anti-phospho-ZAP-70 pTyr315/pTyr319) antibody is specific for human ZAP-70. The product is expected to react with mouse ZAP-70 (as its 100% homologous to human ZAP-70), but has not been tested for it.

Immunogen

synthetic phosphopeptide derived from the region of human ZAP-70 that contains tyrosines 315 and 319.

Application

Anti-phospho-ZAP-70 pTyr315/pTyr319) antibody is suitable for use in western blot (1:1,000 using Jurkat cells treated with H2O2 and immunoblot.

Physical form

Solution in Dulbecco′s phosphate buffered saline (without Mg2+and Ca2+), pH 7.3, with 50% glycerol, 1.0 mg/mL BSA (IgG and protease free) and 0.05% sodium azide.

Accumulating evidence indicates that the interdomain B regions of ZAP-70 and Syk play pivotal roles in the coupling of T-cell antigen receptor (TCR) stimulation to the activation of downstream signaling pathways. The interdomain B region of ZAP-70 contains at least

Following T cell antigen receptor (TCR) engagement, the protein tyrosine kinase (PTK) ZAP-70 is rapidly phosphorylated on several tyrosine residues, presumably by two mechanisms: an autophosphorylation and a trans-phosphorylation by the Src-family PTK Lck. These events have been implicated in

Protein-tyrosine kinases (PTKs) play an integral role in T cell activation. Stimulation of the T cell antigen receptor (TCR) results in tyrosine phosphorylation of a number of cellular substrates. One of these is the TCR zeta chain, which can mediate
