slides can be dried by leaving them at room temperature overnight or by heating them at 37-40 degrees C for several hours. However, it is standard lab policy to wait at least 3 or 4 days before filing the slides, especially if excess mounting media was used. Storing slides in a cabinet before they are fully dry could lead to them sticking together.

Is the organo/limonene mounting media a hard-setting media, or does it require extra sealing on the coverslip (i.e. nail polish)?

This product is a hard-setting mounting medium, sealing is not required. Simply add 2-3 drops of mounting medium and carefully apply the coverslip, avoiding air bubbles. Slides may be dried overnight at room temperature or by incubating at 37-40°C for several hours.

Yes, limonene would be considered a suitable xylene substitute. There are however a wide variety of xylene substitutes available on the market. No exhaustive comparative studies have been performed using Organo/Limonene Mount with each and every available xylene substitute.

Suitable chromogens for immunohistochemistry would be DAB, or DAB with an added metal such as nickel or cobalt. Alkaline phosphatase chromogens would also be suitable for use with Oregano/Limonene Mount.

How does the storage temperature relate to shipping conditions?

The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

What are the instructions on preparing a sample for use with Product No. O8015 (Organo/Limonene Mount™)?

Dehydrate slides with dehydrating agents, such as graded ethanol or reagent alcohol. Dehydration refers to removing the water by using a series of grades alcohols. Typically slides spend 1-5 minutes each in a series of alcohols. Sequences vary by laboratory. A typical sequence would be 1 station of 80% alcohol, 2-3 changes of 95% alcohol, 2-3 changes of absolute alcohol. The slides are then processed through 2-3 changes of xylene. After finishing with the last xylene solution, the slides are ready for coverslipping with the Organo/Limonene Mount.
