General description

The Acetylcholinesterase Activity Assay Kit (Colorimetric) provides a simple, quick, and direct procedure for measuring Acetylcholinesterase levels in a variety of samples such as serum, plasma, tissues or cell lysates / cell extracts. This assay is based on the Ellman method in which thiocholine, produced by Acetylcholinesterase, reacts with DTNB 5,5¢-dithiobis (2-nitrobenzoic acid) to form a colorimetric product (412 nm), proportional to the Acetylcholinesterase activity present. This kit does not require weighing, or mixing of multiple reagents. The Acetylcholinesterase activity is measured without the use of a calibration curve. The Acetylcholinesterase Positive Control in the kit can be used to screen for Acetylcholinesterase inhibitors.

Acetylcholinesterase ( AChE) is a serine hydrolase found at neuromuscular junctions and cholinergic brain synapses. AChE, hydrolyzes choline esters. Acethlcholinesterase activity is related to the function of the nerve response. Its principal biological role is the termination of impulse transmission at cholinergic synapses by rapid hydrolysis of the neurotransmitter acetylcholine (ACh) to acetate and choline.

Organophosphorus (OP) poisons form a covalent bond with a serine residue at the active site of AChE, and are thus potent irreversible inhibitors of AChE.

AChE inhibitors are used in treatment of various neuromuscular disorders, and have provided the first generation of drugs for the treatment of Alzheimer′s disease.

AChE inhibitors inhibit the cholinesterase enzyme from breaking down ACh, increasing both the level and duration of the neurotransmitter action According to the mode of action, AChE inhibitors can be divided into two groups: irreversible and reversible. Reversible inhibitors, competitive or noncompetitive, mostly have therapeutic applications, while toxic effects are associated with irreversible AChE activity modulators.

  2. 1. Is this kit suitable for estimation of red blood cell acetylcholinesterase activity? 2. Can Triton be used for haemolysate preparation?

    1. This product is suitable for assaying acetylcholinesterase activity in cell lysates such as red blood cell hemolysates. Triton X-100 has not been tested for use in this kit. While there are published protocols that do suggest up to 1% Triton X-100, the impact if this detergent on results has not been evaluated. For cell lysates, it is recommended to prepare samples by brief sonication or by homogenization in 0.1 M phosphate buffer (pH 8.0), followed by centrifugation at 14,000 rpm for 5 minutes. The clear supernatant is used for the assay. The Assay Buffer included with the kit should be used for sample dilution.

  3. Is Acetylcholinesterase compatible with the Roche platform, specifically the COBAS 6000?

    1. The product in question, an acetylcholinesterase enzyme or kit, is not compatible with the ROCHE Cobas 6000, as it is a clinical analyzer for clinical chemistry or immunochemistry. All acetylcholinesterase products available are intended for research purposes only and not for human or clinical use.

  4. Is this kit only for acetylcholinesterase or also tests for butylcholinesterase? Is this testing total cholinesterase activity or just AChE activity?

    1. Product CS0003 only measures Acetylcholinesterase not butylcholinesterase and measures AChE activity.

  5. Is product CS0003, Acetylcholinesterase Activity Assay Kit, suitable for use with whole blood? What is the recommended sample preparation protocol, e.g. the proper amount to dilute whole blood for assay?

    1. Yes, kit CS0003 is suitable to test ACHE activity in whole blood. It is recommended to begin with (at least) 1:100 working dilution and progress from there.

  6. What is the difference between the two Acetylcholinesterase Activity Assay Kits MAK119 and CS0003?

    1. Product CS0003 and MAK119 are produced by different manufacturers. In addition, CS0003 is sufficient for 200 tests while product MAK119 is sufficient for 100 tests. The principles and detection range are the same for both products.
      The following is true for product CS0003:
      - Direct procedure for measuring AChE levels in a variety of samples such as serum and plasma.
      - Based on the Ellman method in which thiocholine, produced by AChE, reacts with 5,5'-dithiobis (2-nitrobenzoic acid) to form a colorimetric (412 nm) product, proportional to the AChE activity present.
      - The AChE activity is measured without the use of a calibration curve. The AChE Positive Control in the kit can be used to screen for AChE inhibitors.
      - This kit has a linear range of 10-600 units/liter of AChE activity.
      The following is true for product MAK119:
      - Direct assays of acetylcholinesterase activity in blood, serum, plasma, and other biological samples.
      - Based on an improved Ellman method, in which thiocholine produced by the action of acetylcholinesterase forms a yellow color with 5,5’-dithiobis(2-nitrobenzoic acid). The intensity of the product color, measured at 412 nm, is proportionate to the enzyme activity in the sample.
      - Detection range 10 to 600 U/L AChE activity in 96-well plate assay.
