Application

Adenosine-5′-diphosphoglucose (ADP-Glucose) is a substrate widely used as a glucose donor. It is a substrate in starch synthesis by starch synthase (EC 2.4.1.21). ADP-Glucose is a substrate for alpha-1,4-glucan-protein synthase (ADP-forming) (EC 2.4.1.113), ADP-glucose pyrophosphorylases and many glucosyltransferases.
Adenosine-5′-diphosphoglucose disodium salt has been used as a component of the reaction mixture to measure the enzymatic activity of adenosine-5′-diphosphate (ADP)-glucose pyrophosphorylase (AGPase) and for the synthesis of starch.

ADP-glucose pyrophosphorylase (ADP-Glc PPase) catalyzes the first committed step for the synthesis of glycogen in cyanobacteria and starch in green algae and plants. The enzyme from cyanobacteria is homotetrameric (α4), while that from green algae and plants is heterotetrameric (α2β2).

Solid semi-crystalline starch and hydrosoluble glycogen define two distinct physical states of the same type of storage polysaccharide. Appearance of semi-crystalline storage polysaccharides appears linked to the requirement of unicellular diazotrophic cyanobacteria to fuel nitrogenase and protect it from oxygen
