General description

Osteoprotegerin/TNFRSF11B, also known as Tumor necrosis factor receptor superfamily member 11B, Osteoclastogenesis inhibitory factor, or Osteoprotegerin, (OPG), and encoded by the gene TNFRSF11B/OCIF/OPG, is a membrane bound receptor that acts as a decoy receptor for TNFSF11/RANKL and thereby helps to neutralize or regulate its function in osteoclastogenesis. Bone homeostasis is at least partially dependent upon the levels of TNFSF11 and TNFSF11B, and by acting as decoy or "extra" receptor for TNFSF11B the excess factor levels can be modulated. The activation of the receptors inhibits the activation of osteoclasts and promotes osteoclast apoptosis in vitro. OPG also seems to act as a decoy receptor for TNFSF10/TRAIL and help prevent against TRAIL mediated apoptosis, and TRAIL acts as antagonist for osteoclastogenesis, and thus again OPG appears to moderate osteoclast levels in a dynamic fashion. OPG is highly expressed in numerous tissues other than developing and adult bone germinal centers, but its exact role in these other tissues is still being examined.

Specificity

Immunogen is 92% identical in bovine and in mouse. Immunogen is 84% identical in rat.

KLH-conjugated linear peptide corresponding to the TNFR-Cys 2 domain of human Osteoprotegerin/TNFRSF11B.

Application

Immunohistochemistry Analysis: A 1:500 dilution from a representative lot detected Osteoprotegerin/TNFRSF11B in human kidney tissue.
This Anti-Osteoprotegerin/TNFRSF11B Antibody is validated for use in western blotting & IHC (Paraffin) for the detection of Osteoprotegerin/TNFRSF11B.

Quality

Evaluated by Western Blotting in MG63 conditioned media.

Western Blotting Analysis: 0.2 µg/mL of this antibody detected Osteoprotegerin/TNFRSF11B in 10 µg of MG63 conditioned media.

Target description

~55 kDa observed. Protein is heavily glycosylated and often observed as a monomer at ~55-60 kDa (Cundy T., et al. (2002) Hum Mol Genet. 11(18):2119-27).
