Descripción general

La insulina es una hormona polipeptídica bicatenaria producida por las células β de los islotes pancreáticos. Su peso molecular es de ~ 5800 Da) Las cadenas α y β se unen por dos enlaces disulfuro intercatenarios. La cadena α contiene un puente disulfuro intracatenario.

Aplicación

La disolución de insulina humana se ha utilizado como suplemento en el medio de Eagle modificado de Dulbecco (DMEM) para cultivar la línea de células epiteliales y la línea de células endometriales humanas, el medio de Eagle esencial mínimo para cultivar la línea de células de adenocarcinoma de mama humanas y el medio 199 para cultivar la línea celular ovárica sana.
La disolución de insulina humana se recomienda para aplicaciones de cultivo celular a 0,5 -1ml por litro de medio.
La disolución de insulina humana se ha utilizado para la prueba de tolerancia a la insulina en ratones y ratas.[1][2] También se ha utilizado para la diferenciación adipogénica de cultivos de células madre embrionarias humanas (hESC) y hMSCs (células del estroma mesenquimatoso humano).[3][4]

Acciones bioquímicas o fisiológicas

La insulina desempeña un papel en varios procesos celulares, como la promoción del crecimiento celular, la regulación del ciclo celular y la facilitación de la captación de glucosa. Suele utilizarse como suplemento proteico en medios carentes de suero para cultivo de células de mamíferos con el objetivo de contribuir a promover el crecimiento y el mantenimiento óptimos de las células de mamíferos.

    1. For research applications, freezing insulin solution has been a common pratice. However, solutions available on the commercial market for therapeutic use are never frozen, in order to maintain maximum activity.
