生化/生理作用
TVB-3166是一种可渗透细胞的吡唑衍生物,以有效和可逆方式(IC50 = 42 nM)通过脂肪酸合酶(FASN)酮还原酶活性,对细胞棕榈酸酯的合成进行抑制(IC50 <100 nM) ,同时有效抑制培养物中各种癌细胞的棕榈酸酯依赖性存活和体内小鼠异种移植肿瘤的生长(60-100 mg/kg /d p.o.),但对癌症和非癌症培养物的非棕榈酸酯依赖性生长几乎没有影响或没有抗增殖活性。TVB-3166处理对FASN的抑制作用还对培养物中(有效浓度为200 nM,靶向A549、HEp2和HeLa培养物中的RSV和PIV3)和小鼠(50 mg/kg/12 h,p.o.;RSV)体内 的复制呼吸道病毒有效,并对宿主细胞或小鼠无明显不利影响。
