淀粉样蛋白-β(Aβ)肽是阿尔茨海默病大脑的老年斑特征的主要成分。它是一种I型跨膜蛋白。编码淀粉样蛋白-β的基因定位于人21号染色体上。
應用
本品人淀粉样前体蛋白β已用作肽标准品。
生化/生理作用
分泌型β淀粉样蛋白前体(sAPPβ)是β淀粉样蛋白前体(APP)水解得到的裂解产物。APP被β-分泌酶切成2个片段,sAPPβ和β-淀粉样肽(Aβ)。Aβ是阿尔茨海默病(AD)的主要标志之一——淀粉样老年斑的成分,sAPPβ被认为可调节神经元功能和细胞存活。瑞典家系AD中存在的APP突变使APP更易被β-分泌酶切割,形成更多的Aβ和sAPPβ。相反,蛋白激酶C(PKC)激活可降低sAPPβ浓度。反面高尔基体网状结构(TGN)是β-分泌酶活性发挥的主要位点。
