烯醇化酶是一种多功能糖酵解酶,催化 D -2-磷酸甘油酸转化为磷酸(烯醇)丙酮酸 (PEP) 和水。它可作为内皮、上皮和造血细胞中的纤溶酶原受体,因此可能参与纤维蛋白溶解和血管内系统。它还被认为是一种热休克蛋白,可能在转录和病理功能中起到作用。烯醇化酶与自身免疫和系统性疾病有关。此外,已知血清神经元特异性烯醇化酶可作为心脏骤停后患者预后的预测因子。因此,烯醇化酶分析可用于研究细胞功能,如碳水化合物代谢、转录和其他病理生理过程。

原則
通过偶联酶试验测定烯醇酶活性,其中 D-2-磷酸甘油酸转化为 PEP,形成中间体与过氧化物酶底物反应,生成比色 (570 nm) 或荧光(λex = 535/λ em = 587 nm)产物与存在的烯醇化酶活性成正比。一毫单位烯醇化酶是 25°C、pH 7.2,每分钟生成 1.0 nmol H 2 O 2 的酶量。
