Inflammation and immune cell response are key to maintaining homeostasis. Inflammation plays a central role in the etiology of many immunologically related diseases, such as infectious disease, osteoarthritis and respiratory disease, as well as neurologic, metabolic and cardiovascular disease and even cancer. However, until now the ability to measure levels of cytokines and chemokines in sheep samples has been limited primarily to ELISAs. Researchers now have the ability to measure multiple inflammation biomarkers simultaneously using our MILLIPLEX® Ovine Cytokine/Chemokine Panel 1, based on the Luminex® xMAP technology.MILLIPLEX® Ovine Cytokine/Chemokine Panel 1 may be used for the simultaneous quantification of any or all of the following analytes in serum, plasma, and tissue culture supernatant samples: IFNγ, IL-1α, IL-1β, IL-4, IL-6, IL-8 (CXCL8), IL-10, IL-17A, IL-36RA (IL-1F5), IP-10 (CXCL10), MIP-1α (CCL3), MIP-1β (CCL4), TNFα, VEGF-A. Note: The protocol recommends a 1:2 dilution of serum and plasma samples, except when assaying IL-8 (CXCL8) in serum samples for which an up to 1:8 dilution of sample may be required.MILLIPLEX® offers the broadest selection of analytes across a wide range of disease states and species. Once the analytes of interest have been identified, you can rely on the quality that we build into each kit to produce results you can trust. In addition to the assay characteristics listed in the protocol, other performance criteria evaluated during the verification process include: cross-reactivity, dilution linearity, kit stability, and sample behavior (e.g. detectability and stability). In addition each panel and kit meets stringent manufacturing criteria to ensure batch-to-batch reproducibility.Each MILLIPLEX® panel and kit includes:Quality controls (QCs) to qualify assay performanceComparison of standard (calibrator) and QC lots to a reference lot to ensure lot-to-lot consistencyOptimized serum matrix to mimic native analyte environment in serum and plasma samplesDetection antibody cocktails designed to yield consistent analyte profiles within panelThe MILLIPLEX® Ovine Cytokine/Chemokine Panel 1 thus enables you to focus on the therapeutic potential of cytokines and the modulation of cytokine expression.

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

应用

The Ovine Cytokine/Chemokine bead-based multiplex cytokine panel, using the Luminex® xMAP technology, enables the simultaneous analysis of 14 cytokine and chemokine biomarkers in ovine serum, plasma and cell culture samples.This is an overnight incubation assay.The protocol recommends a 1:2 dilution of serum and plasma samples, except when assaying IL-8 (CXCL8) in serum samples for which an up to 1:8 dilution of sample may be required.This kit may be used for the analysis of any or all of the Ovine Cytokine/Chemokine Panel 1 analytes in ovine tissue/cell lysate and culture supernatant samples and serum or plasma samples.Analytes available:IFN-γ;IL-1α;IL-1β;IL-4;IL-6;IL-10;IL-8;IL-17A;MIP-1α;IL-36RA;IP-10;MIP-1β;TNFα;VEGF-A

  1. Can CSF samples be utilized in a Milliplex kit that does not explicitly mention CSF samples in the protocol?

    1. Cerebrospinal fluid (CSF) samples can be used in Milliplex kits if not mentioned in the protocol, with the caution that it is not validated. The following steps are recommended for processing CSF samples:
      - Collect and centrifuge samples to remove blood cells.
      - Avoid using pink samples that may indicate breakdown of blood cells.
      - Spin at high speed for 10 minutes in a benchtop microcentrifuge.
      - Collect the top clear portion of the samples, aliquot, and freeze at -80°C.
      - Use Assay Buffer as the matrix.
      - Test a few dilutions to find out the optimal sample dilution factor.

  2. Do the Bovine Cytokine and Ovine Cytokine Milliplex kits utilize the same antibody pairs?

    1. The kits utilize the same capture and detection antibodies for all cytokines, with the exception of IL-8.
