干扰素-γ(IFN-γ)是由活化的NK细胞和CD4+TCRα/β+,CD8+TCRα/β+和TCRγ/δ+T细胞产生的调节蛋白。 IFN-γ的生物活性包括与一类高亲和力受体的特异性结合。 此外,IFN-γ可以升级淋巴样细胞功能的数量,包括巨噬细胞,NK细胞和中性粒细胞的抗微生物和抗肿瘤反应。重组小鼠IFN-γ是一种含134个氨基酸的蛋白,预测分子量为15.6 kDa。在大肠杆菌发酵过程中由于蛋白水解裂解出现C末端截短,并产生一种经质谱验证为15.1 kDa的蛋白。重组小鼠IFN-γ的SDS-PAGE 分析出现一条11-12 kDa的条带,在非还原和还原条件下,该条带在11-12 kDa左右迁移。
應用
对于大多数体外应用,IFN-γ在0.1至10.0 ng/mL的浓度范围内发挥其生物活性。

外觀
10 mM磷酸钠、pH值7.0+25 mM NaCl经无菌过滤、冻干。
儲存和穩定性
尽管冻干的蛋白质在室温下可稳定数周,但最好在 -20°C 下保存。快速旋转后,用磷酸钠,pH值8.0复溶至1 mg/mL的浓度。 进一步稀释应配成RPMI+10%FCS或5%HAS在2-8ºC保存一周或在-20ºC保存备用。自收到之日起,复溶的IFN-γ应该以未稀释的等分试样在 -20°C 下保存长达 6 个月。
